expression of Haematococcus pluvialis peroxiredoxin genes under different culture conditions.
초록
The unicellular green alga, Haematococcus pluvialisis known for its massive accumulation of the astaxanthin in cytocol. Astaxanthin is a kind of carotenoid as an antioxidant and synthesized in various environmental stress such as strong irradiance or nitrogen deficiency. Organisms that containing high level of astaxanthin are shrimps, salmons etc. And astaxanthin is used source o f pigmentation for fish aquacultures, cosmetics and as a potential drug in cancer prevention as a free-radical quencher. Peroxiredoxin (Prxs) are thiol-specific antioxidant protein. Prxs exert their protective antioxidant role in cells through their peroxidase activity (ROOH + 2e- -> ROH + H2O), whereby hydrogen peroxide, peroxynitrate, and a wide range of organic organic hydroperoxides (ROOH) are reduced and detoxified. In this study, we compared with samples under each environmental stress with HPLC. And we discovered some peroxiredoxin genes that are related with synthesis of astaxanthin through cDNA library and then classified the prx genes. Among these prx genes, we choice one prx gene expected to Prx Q and then we confirmed copy number of gene by southern blot analysis. And we make a comparative study for expression level of prx gene in various stress condition. Results of this study will be discussed.
