Use of Ascrobic Acid and Its Derivative as Supplements for Mouse Fibroblast Culture in Medium with Reduced Serum

In this study, we investigated the effects of ascorbic acid and its derivative on the proliferation of 3T3-J2 mouse fibroblasts in medium with reduced serum. The mouse fibroblast feeder layer enables human keratinocytes to have colony-forming potential without the terminal differentiation for in-vitro reconstitution of skin. In order for cultured keratinocytes to become a clinical application, it is necessary that stable and rapid expansion of fibroblasts in defined culture conditions. We considered ascorbic acid as an essential supplement for developing serum-free media. Ascorbic acid has not only the antioxidant effect on intracellular reactive oxygen species but it also stimulates the production of collagen. On the other hand, ascorbic acid is a very unstable component in aqueous solutions. It can easily decompose in the culture medium. Moreover, it has studied that there are antiproliferative effects of ascorbate in millimolar concentrations. To circumvent the drawbacks of ascorbic acid, we examined the possibility of using an ascorbic acid derivative for replacing ascorbic acid and increasing proliferation.
