SNP of Phospholipase D1 Gene as a predictive marker for pro-cancer detection

Phospholipase D (PLD) is activated in response to stimulators of vesicle transport, endocytosis, exocytosis, cell migration, and mitosis. These cellular biological processes have been known to be deregulated in the development of various human tumors. However, There was no investigation to figure out the role of PLD single nucleotide polymorphisms (SNPs) which can be extensively used in case-control studies of practically all cancer types. The purpose of this study was to analyze genetic polymorphisms of Phospholipase D1 (PLD1) gene as a predictive marker for pro-cancer detection. We carried out analysis of SNP of this gene by PCR direct sequencing after DGGE. A heterozygous C to A transition at the 74th base in Exon 23 which was novel among 26ea exons of this gene has been in Lung cancer and colorectal carcinoma patients. The allelic frequency of this gene was 62.2% (79 cases, p<0.005) in lung cancer 127 cases and 88.46% (46 cases, p<0.005) in colon cancer 52 cases. Comparatively, the allelic frequency of exon23-2699bp-allele A/C was 16.85% (15 cases) in normal 89 cases. From our results, We suggest that PLD1 genetic polymorphism might be a predictive marker for pro-cancer detection. Further studies with large number of cases will be needed.
