Adjustment of Inorganic Salts Concentrations for Mouse Fibroblast Culture in Serum-Free Medium

Inorganic salts, contained in the serum-free medium, are heavy metals which work as trace elements and buffer substances and so on, being essential to cell proliferation. In spite of its importance however, consideration about inorganic salts are not established yet. The reason why consideration about inorganic salts is important is that trace elements can be toxic to the cells in serum-free medium, which does not contain serum. In case of human keratinocytes, inorganic salts like Zn, Cu, Mn etc. give beneficial effect to the proliferation when added in extremely small quantities. But it has been reported that when it was provided in excess, it was toxic to cell proliferation instead. So we cultivated fibroblasts in commercial media, regulating the inorganic salts concentrations. The medium was identical, the cell growth of the mouse fibroblast was lower than the cell growth of the human fibroblast. We assumed the reason of the mouse fibroblast low growth was that mouse fibroblast was more sensitive to inorganic salts concentrations than human fibroblast. So in this study, we seeked for the adequate inorganic salts concentration, in order to develop a serum-free media practicable for mouse fibroblast growth.
