Differential Expression of genes at implantation sites compared to interimplantation sites in mouse uterus using cDNA Microarray

The process of embryo implantation involves complex interactions between the blastocyst and the uterus. Successful implantation requires not only growth of the blastocyst, but also establishment of receptive uterus. While it is well established that appropriate maternal steroid hormones are essential for endometrial preparation for implantation, but the molecular events at the actual site of implantation are still little understood. Our aim was to identify and characterize genes uniquely regulated at the site of implantation in mouse uterus by cDNA microarray. cDNA microarray was performed for implantation site and interimplantation site in mouse uterus Day 4.5 of pregnancy (plug check = Day 1). Of the 5355 genes analyzed, 130 genes increased about two-fold level of expression in the implantation site and 192 genes decreased about two-fold level of expression in the implantation site. Candidate genes analyzed expression patterns in the implantation site compared to interimplantation site using reverse-transcription polymerase chain reaction (RT-PCR). This finding suggest that activation and deactivation of these genes are required for successful implantation, and may be to aid our understanding on the nature of the implantation.
