Mammalian cells have specific requirements for cholesterol, and it is recognized as an essential nutrient in cell growth.1) Many cell types possess mechanisms to internalize exogenous sources of cholesterol, but most cells satisfy their requirements by endogenous cholesterol biosynthesis.2) While cholesterol is necessary in cell culture, addition of cholesterol in the culture media depends on a case by case basis. Reports have informed inclusion of cholesterol in the culture media has resulted in diverse effects to various cell lines, displaying multiplicity to proliferation and end-product production. An investigation about how the addition of cholesterol to a culture media affects the cell growth should be necessary. We added cholesterol to a commercial serum-free media via cyclodextrin-cholesterol complexes. An observation of the appearances of cell growth for suspension culture was conducted. The cultivation conditions were performed on various commercial serum-free media and various cells. The cells were all an identical CHO-DG44 cell line, but originated from a different selection route. Insertion of cholesterol leaded to numerous results which differed depending on the cell culture medium and the cell line. Remarkable improvements for proliferation were observed on certain conditions. The results indicate that addition of cholesterol to culture media may be required depending on type of the cell and the media.
