초록
In the preceding study, we observed the effects of ascorbic acid derivatives, a replacement of ascorbic acid, on the mouse fibroblast. By adding ascorbic acid derivatives in the mouse fibroblast cell, we found out that the cells survived for a longer period. CHO cell, used most widely in recombinent protein production, is required a longevity for the more products. A recent study has shown that L-ascorbic acid 2-phosphate, an ascorbic acid derivative having anti-oxidant functions, inhibited apoptosis in suspension culture of the CHO cell. 1) In case of ascorbic acid as an anti-oxidant, it"s very oxidative and unstable in aqueous solution. Since it"s easily decomposed in the culture medium, we put forth the need of the development of the media containing ascorbic acid derivatives.2) So in this study, we looked at whether ascorbic acid derivatives, showing efficacy in previous studies, could be useable in reduced serum medium or serum free medium for the non-adapted CHO cell. Further more, by investigation on the effect on CHO cells adapted for suspension culture, we looked over the possibility of the development of a suspension culture media containing ascorbic acid derivatives
