Effect of Haloperidol and Risperidone Treatment on HSP70 Expression by MK-801 in Rat Glioma Cells

The aim of this study is to evaluate the differences in the effects of antipsychotics, haloperidol and risperidone, on expression of HSP70 and the relationship between antipsychotics and NMDA receptor. This study investigated heat shock protein 70 (HSP70) expression after haloperidol and risperidone treatment on rat C6 glioma cell. To examine how the MK-801 affects the extent of HSP70 expression in nerve cells, the rat C6 glioma cells were treated with MK-801, a neurotoxin, for 6 hours, and the extent of HSP70 expression was examined using anti-HSP70 monoclonal antibody by HSP70 immunochemistry and Western blots technique. And, to experiment schizophrenia-like condition, haloperidol or risperidone was administered to the experimental groups of C6 glioma cells which include MK-801 (10uM) which causes schizophrenia-like symptomsfor 1, 6, 24, and 72 hours each, after which the extentof HSP70 expression was investigated and compared. Finally, haloperidol treatment on C6 glioma cell significantly decreased the level of HSP70 expression compared with before treatment. In addition, risperidone treatment groups also showed decreased expression of HSP70 as the duration of risperidone treatment became longer. This study suggests mechanism that haloperidol and risperidone could have antipsychotic effects by decreasing HSP70 expressionunder the effect of stress such as MK-801.
