Transcriptome analysis based on microarray in Chaetoceros neogracile

Transcriptome analysis based on microarray in Chaetoceros neogracile Antarctic diatom Chaetoceros neogracile is an important organism in understanding environmental stress including cold temperature. The growth curve clearly shows that irradiance of 600μE·m-2·s-1 and temperature of 10℃ inhibit the growth of C.neogracile, and we decided this condition to be high light and heat stress respectively. We previously constructed and analyzed EST library from this diatom to to gain insights on the transcriptomes of this Antarctic diatom. In this study, a customized cDNA chip was hybridized with RNA sample isolated under light and heat stress condition. Among 1439 probes, 40% were differentially regulated (>2-fold) under the high irradiance, and 21% under the heat stress. The pigment profile was analyzed by HPLC shows xanthophyll cycle in this diatom.
